An Image Analytical Study of Acute Lymphoblastic Leukemia Cells


Abstract
FAB classification of acute lymphoblastic leukemia(ALL) is of significance for prognosis, but diagnostic disagreement between pathologists may be considerable due to the qualitative and subjective nature of the assessment by FAB criteria. In
order
to
investigate the adjuvant potential of image analysis in classification and prognosis of ALL, the present study was undertaken. Using and image analyzer(IBAS 2000, Kontron, FRG), the morphologic characteristics and DNA Ploidies of leukemic cells
were
determined in 44 patients with acute lymphoblastic loukemia(FAB-L1, L2) and clinical follow-up studies were done for more than 12 month The mean number, standard deviation of nucleoli and nuclear from factor were important variables in
discriminating
the FAB subgroups(FAB-L1, L2). The overall agreement between subjective FAB classification and the grouping predicted by the morphometric data was 88.9%. On morphometric data, longer perimeter of nucleus, larger number of nucleoli and more
irregular
shape of nucleus were associated with short-term survival. Aneuploidy was found in 15 cases(34.1%) in ALL(FAB-L1, L2). However, DNA ploidy patterns cstimated by image analyzer didn't have any relation to survival duration of ALL.
